Affinity chromatography is a method of separating biochemical mixtures based on highly specific biological interactions. The Glutathione Agarose is developed for purification of fusion-proteins expressing a Glutathione S-transferase (GST) moiety. This GST-glutathione interaction makes one-step purification possible for proteins under native conditions. GST has a high affinity for its substrate, glutathione, which has been covalently bound to a solid support. Impurities are removed and the bound GST-fusion proteins can be eluted by using buffers containing reduced glutathione. Applications Purification of Glutathione-S-transferase (GST) and GST-tagged fusion proteins. Antibody purification. Assays involving protein-protein and protein-DNA interactions. Structural and functional investigation of proteins. Special Features One-step easy purification protocol from crude lysate. Purification under native conditions. Can be used for proteins with low expression. Can be used for successive cycles. Ready-to-use for any scale of purification. Protein binding capacity: approx. > 8mg recombinant GST/ml gel. Resins and cartridges formats suitable for MPLC, FPLC, AKTA design, batch, gravity, peristaltic pump and syringe. Beads are supplied as 75% (V/V) aqueous suspension (in 20% ethanol). Download Protocl
